Biological Background: RRM2B Function and Localization
- RRM2B (p53R2) is a TP53-inducible ribonucleotide reductase subunit that plays a pivotal role in DNA damage repair by supplying deoxyribonucleotides in a p53-dependent manner.
- It forms an active ribonucleotide reductase (RNR) complex with RRM1, which is expressed in both resting and proliferating cells upon DNA damage.
- The enzyme contains an iron-tyrosyl free radical center essential for catalytic activity, classifying it as an iron-binding oxidoreductase.
- Subcellular localization is detected in both the cytoplasm and nucleus, consistent with its role in DNA repair processes.
- Tissue expression is widespread, with high levels in skeletal muscle, weak expression in thymus, and positive staining reported in epithelial dysplasias and squamous cell carcinomas.
- Genetic variants in RRM2B are associated with mitochondrial DNA depletion syndromes, deafness, neuropathy, and progressive external ophthalmoplegia.
Experimental Guidance and Technical Tips
- The immunogen comprises a synthetic peptide derived from the N-terminal region (amino acids 1–100) of human RRM2B; consider this epitope location when designing experiments.
- This antibody is recommended for ELISA and immunofluorescence/immunocytochemistry (IF/ICC) applications as indicated in the specifications.
- Optimal working dilutions must be determined empirically in the end-user’s own experimental system.
- For IF/ICC, optimize fixation and permeabilization protocols to detect both cytoplasmic and nuclear pools of RRM2B.
- As a polyclonal reagent, it may recognize multiple epitopes; include appropriate controls to confirm specificity in your model.
