Biological Background: RRAS2 Function and Localization
- RRAS2, also known as Ras-like protein TC21 or teratocarcinoma oncogene, is a member of the Ras superfamily of small GTPases encoded by the RRAS2 gene. Alternative splicing produces multiple isoforms.
- It functions as a GTP-binding protein with intrinsic GTPase activity, playing a central role in the regulation of the MAPK signaling pathway and thereby controlling diverse cellular processes. Related references: PMID:31130282 PMID:31130285 PMID:39809765
- RRAS2 is critically involved in craniofacial development, as demonstrated by genetic studies linking its function to morphogenetic events during embryogenesis. Related references: PMID:31130282 PMID:31130285
- At the subcellular level, RRAS2 localizes primarily to the cell membrane and the Golgi apparatus membrane, consistent with its roles in signal transduction and vesicular trafficking.
- It is ubiquitously expressed across human tissues, with the highest levels detected in heart, placenta, and skeletal muscle, moderate expression in lung and liver, and lower levels in brain, kidney, and pancreas.
- Post-translational modifications of RRAS2 include palmitoylation, prenylation, phosphorylation, acetylation, and methylation, which modulate its membrane association, activity, and interactions.
- Classified as a proto-oncogene, aberrant activation or overexpression of RRAS2 has been implicated in oncogenic transformation and is associated with disease variants in certain cancers.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human RRAS2 (amino acids 1-204), which may confer broad isoform reactivity; consider verifying specificity in your experimental system.
- For Western blot, the predicted molecular weight is approximately 23 kDa; use appropriate gel percentages and membrane transfer conditions for low-molecular-weight proteins.
- As a polyclonal antibody, lot-to-lot consistency may vary; include a positive control (e.g., HEK293 or HeLa lysates) and validate performance before critical experiments.
- ELISA applications may require optimizing coating antigen concentration and antibody dilution; recombinant protein immunogen can serve as a positive control.
