Biological Background: RRAGA Function and Localization

  • RRAGA (Ras-related GTP-binding protein A), also known as FIP-1 or RAGA, is a member of the Rag GTPase family.
  • Guanine nucleotide-binding protein that plays a crucial role in the cellular response to amino acid availability through regulation of the mTORC1 signaling cascade. RRAGA forms heterodimeric Rag complexes with RagC/RRAGC or RagD/RRAGD and cycles between an inactive GDP-bound and an active GTP-bound form.   • In its GTP-bound active form, RRAGA promotes the recruitment of mTORC1 to the lysosomes and its subsequent activation by the GTPase RHEB. Related references: PMID:31601708

  • For Western Blot, the predicted molecular weight is approximately 37 kDa, though UniProt annotates 36.6 kDa. Validate in the relevant sample system and consider loading controls.
  • For IHC-P, tissue sections from skeletal muscle, heart, or brain are recommended for positive staining based on expression data. Optimization of antigen retrieval and antibody dilution is advised.
