Biological Background: RPS6KA5 (MSK1) Function and Localization
- RPS6KA5, also known as MSK1 or ribosomal protein S6 kinase alpha-5, is a serine/threonine-protein kinase and a member of the RSK family. Alternative splicing produces isoforms with predicted molecular weights of 62, 82, and 90 kDa.
- Phosphorylates transcription factors CREB1 and ATF1 in response to mitogenic and stress stimuli (e.g., UV-C, EGF, anisomycin), driving immediate-early gene expression. Related references: PMID:11909979, PMID:9873047
- Mediates RELA/NF-κB phosphorylation at Ser-276, critical for TNF-induced transcriptional activity and glucocorticoid-mediated inflammatory gene repression. Related references: PMID:12628924, PMID:18511904
- Phosphorylates histone H3 at Ser-10 and Ser-28, and histone H2A at Ser-1, contributing to chromatin remodeling and transcriptional activation of proto-oncogenes c-FOS and c-JUN. Related references: PMID:12773393, PMID:15010469
- Also phosphorylates STAT3 (Ser-727) in erythropoietin-stimulated cells, ETV1/ER81 (Ser-191, Ser-216) regulating transcription in development and tumors, and TRIM7 (Ser-107) to stimulate ubiquitin ligase activity. Related references: PMID:12763138, PMID:12569367, PMID:25851810
- Subcellular localization is both nuclear and cytoplasmic, consistent with functions in transcription regulation and signal transduction.
- Widely expressed with highest levels in heart, brain, and placenta, and lower in lung, kidney, and liver.
- Classified as a stress-response and inflammatory kinase; post-translationally regulated by phosphorylation and ubiquitination.
Experimental Guidance and Technical Tips
- For Western blotting, consider that multiple isoforms (62, 82, 90 kDa) may be detected; optimize antibody dilution and use appropriate molecular weight markers.
- ELISA can be performed with coated antigen or capture antibody; ensure the immunogen sequence (aa 639-736) is present in the target protein.
- Cross-reactivity has been observed with human, mouse, rat, and monkey, but validation in your specific experimental system is recommended.
- The antibody is raised against a synthetic peptide; thus, it may not recognize all alternatively spliced isoforms—confirm specificity using relevant knockdown or overexpression models.
