Biological Background: RPS6KA1/RPS6KA2/RPS6KA3 Function and Localization
- RPS6KA1 (RSK1) is a serine/threonine-protein kinase belonging to the p90 ribosomal S6 kinase (RSK) family, which also includes RPS6KA2 (RSK3) and RPS6KA3 (RSK2).
- Acts downstream of ERK/MAPK signaling to phosphorylate and activate transcription factors such as CREB1, ETV1/ER81, and NR4A1/NUR77, mediating mitogenic and stress-induced gene expression. Related references: PMID:10679322, PMID:12213813, PMID:18508509, PMID:18813292
- Regulates translation by phosphorylating ribosomal protein S6 (RPS6) via an mTOR-independent mechanism and by phosphorylating EIF4B, enhancing cap-dependent translation initiation. Related references: PMID:17360704, PMID:16763566
- Modulates the mTOR nutrient-sensing pathway by directly phosphorylating TSC2 and RPTOR, thereby regulating mTORC1 activity. Related references: PMID:15342917
- Promotes cell survival by phosphorylating and inhibiting pro-apoptotic proteins BAD and DAPK1, and contributes to hepatic stellate cell survival via CEBPB phosphorylation. Related references: PMID:10679322, PMID:16213824, PMID:11684016
- Regulates cell cycle progression by phosphorylating CDKN1B, which promotes its cytoplasmic retention and prevents G1/S inhibition. Related references: PMID:18508509, PMID:18813292
- Controls cell migration through phosphorylation of EPHA2 at Ser-897. Related references: PMID:26158630
- Localizes to both the nucleus and cytoplasm, consistent with its roles in signal transduction and gene regulation.
- During Kaposi's sarcoma-associated herpesvirus infection, constitutively active RPS6KA1 promotes the late transcription and translation of viral lytic genes.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from a conserved region (aa 600–700) shared among RPS6KA1, RPS6KA2, and RPS6KA3, which likely contributes to the antibody's pan-RSK reactivity; consider verifying isoform specificity in your system if needed.
- This rabbit polyclonal antibody is suitable for denaturing (WB) and native (IHC-P, IF/ICC) applications; each assay should be optimized with appropriate positive and negative controls.
- Reported cross-reactivity includes human, mouse, and rat; alignments of the immunogen sequence across species can guide predictions for additional species.
- Polyclonal antibodies may exhibit lot-to-lot variability; validation in the relevant sample system is recommended.
