Biological Background: RPS17 Function and Localization

• RPS17 (40S ribosomal protein S17) is a component of the small ribosomal subunit, which is a large ribonucleoprotein complex responsible for protein synthesis in the cell. Related references: PMID:23636399
• It is part of the small subunit (SSU) processome, the first precursor of the small eukaryotic ribosomal subunit. During assembly in the nucleolus, many ribosome biogenesis factors, an RNA chaperone, and ribosomal proteins associate with nascent pre-rRNA to mediate RNA folding, modifications, rearrangements, cleavage, and targeted degradation of pre-ribosomal RNA by the RNA exosome. • Subcellularly, RPS17 localizes to the cytoplasm and the nucleolus.
• Mutations in RPS17 are associated with Diamond-Blackfan anemia, a congenital erythroid aplasia.
• RPS17 is a phosphoprotein and undergoes ubiquitination (Ubl conjugation), as indicated by proteomics studies.
• It is also known as eS17, and the gene is designated RPS17 (RPS17L).

Experimental Guidance and Technical Tips

• This polyclonal antibody, raised against an N-terminal peptide (aa 1-100), may recognize the full-length RPS17 protein. For western blot, an expected band of approximately 16 kDa should be monitored; validate in the relevant sample system.
• Given the cross-reactivity with mouse and rat, consider using lysates from these species as positive or comparative controls in WB.
