Biological Background: RPRD1B Function and Localization
- RPRD1B (also known as CREPT, Cell cycle-related and expression-elevated protein in tumor) is a nuclear protein that regulates transcription by interacting with the phosphorylated C-terminal domain (CTD) of RNA polymerase II subunit POLR2A.
- It participates in CTD dephosphorylation via RPAP2 and enhances expression of CCND1 (cyclin D1) by promoting RNA polymerase II binding to the CCND1 promoter.
- RPRD1B prevents RNA polymerase II from reading through the 3' end termination site, and may facilitate its recycling to the promoter through chromatin loop formation.
- It also upregulates other cell cycle-related genes such as CDK2, CDK4, CDK6, and cyclin E, while not affecting CDKN1A, CDKN1B, or cyclin A.
- Localized exclusively in the nucleus, RPRD1B is a phosphoprotein and acetylated protein, as per proteomics identifications.
- Preferentially expressed in a variety of tumor tissues (colon, lung, liver, breast, prostate, stomach, uterine endometrium, cervical cancers) with higher levels than adjacent non-tumor tissue, suggesting a role in proliferation and oncogenesis.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 80–326 of human RPRD1B; the polyclonal antibody may recognize multiple epitopes within this region. For positive controls, consider using full-length recombinant protein or lysates from human, mouse, or rat cell lines.
- In Western blotting, a band near 37 kDa is expected. Validate specificity with appropriate blocking and detection conditions, and include molecular weight markers.
- For IHC-P, antigen retrieval may be required. Test on known RPRD1B-overexpressing tumor tissues (e.g., colon or breast cancer) and include a negative control to confirm staining specificity.
