Biological Background: RPN1 Function and Localization

  • RPN1, also known as ribophorin I (RPN-I) or dolichyl-diphosphooligosaccharide--protein glycosyltransferase 67 kDa subunit, is a core subunit of the oligosaccharyl transferase (OST) complex.
  • The OST complex catalyzes the initial transfer of a defined glycan (Glc3Man9GlcNAc2) from dolichol-pyrophosphate to asparagine residues within Asn-X-Ser/Thr consensus motifs, the first step in protein N-glycosylation (PMID:31831667). Related references: PMID:31831667
  • N-glycosylation occurs cotranslationally, and the OST complex associates with the Sec61 translocon complex at the endoplasmic reticulum (ER) membrane.
  • RPN1 is an integral membrane protein localized to the ER membrane and has also been identified in melanosomes.
  • It is expressed in all tissues tested, indicating a ubiquitous role in glycoprotein synthesis.

  • For Western blot, the predicted molecular weight is 68 kDa. Use reducing conditions and appropriate loading controls. Validate with positive control lysates from human, mouse, or rat cell lines (e.g., HeLa, HEK293, NIH/3T3).
  • In ELISA, this antibody may be used for direct detection or capture of recombinant RPN1 protein; consider testing against the immunogen sequence or full-length protein.
  • Cross-reactivity has been observed with mouse and rat RPN1 by supplier validation, but it is advisable to confirm species-specific performance in your experimental system.
  • When using for detection of endogenous RPN1, note that the protein is widely expressed; however, expression levels may vary. Optimization of antibody dilution may be necessary.
