Biological Background: RPN1 Function and Localization
- RPN1 (ribophorin I) is a subunit of the oligosaccharyl transferase (OST) complex, which catalyzes the transfer of a high-mannose glycan from dolichol-pyrophosphate to asparagine residues, initiating protein N-glycosylation. Related references: PMID:31831667
- N-glycosylation occurs cotranslationally as the OST complex associates with the Sec61 translocon, ensuring efficient modification of nascent polypeptides entering the endoplasmic reticulum (By similarity).
- RPN1 is an integral membrane protein of the endoplasmic reticulum and is also detected in melanosomes.
- It is ubiquitously expressed across all tissues tested.
- The protein undergoes post-translational modifications including acetylation and N-glycosylation, and contains transmembrane domains, consistent with its membrane localization.
- Keywords also indicate the presence of a signal sequence, isopeptide bonds, and ubiquitin conjugation, suggesting regulatory mechanisms.
- The gene symbol RPN1 stands for ribophorin I, also known as OST1 or RBPH1.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein spanning amino acids 20–280 of human RPN1, targeting the N-terminal portion of the protein.
- For Western blotting, consider using whole cell lysates from human or rat cell lines, given the ubiquitous expression of RPN1; a band at approximately 69 kDa is expected.
- In ELISA applications, the polyclonal nature may provide robust detection; however, validate with the immunogen or a fragment to ensure specificity.
- Due to high sequence conservation, cross-reactivity with other species is possible; pilot studies are recommended if using the antibody in non-validated species.
