Biological Background: RPL23A Function and Localization
- RPL23A encodes the large ribosomal subunit protein uL23, also known as 60S ribosomal protein L23a, a core component of the 60S ribosomal subunit.
- It binds a specific region on the 26S rRNA, contributing to the structural integrity and peptidyl transferase activity of the large ribosomal subunit during protein synthesis. Related references: PMID:23636399, PMID:32669547
- RPL23A may play a role in regulating p53/TP53 stability by promoting MDM2-mediated polyubiquitination and subsequent proteasomal degradation, linking ribosomal function to cell cycle and apoptosis control. Related references: PMID:26203195
- The protein localizes predominantly to the cytoplasm as part of the ribosomal complex, but it is also found in the nucleus, consistent with its roles in ribosome biogenesis and nuclear-transported functions.
- Post-translational modifications of uL23 include acetylation, citrullination, methylation, phosphorylation, and ubiquitination, which may modulate its assembly into ribosomes or its extraribosomal functions.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein corresponding to the full-length human RPL23A (aa 1-156). When designing detection assays, ensure the epitope is accessible in your target sample's native conformation.
- For Western blot (WB), the predicted molecular weight is ~18 kDa; consider using a suitable percentage of SDS-PAGE gel for low-molecular-weight protein separation and verify with appropriate positive controls (e.g., HeLa, HEK293 cell lysates).
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), validate the antibody in your specific tissue or cell system, as antigen retrieval and fixation methods can affect staining.
