Biological Background: RPL17 Function and Localization
- RPL17, also known as 60S ribosomal protein L17, L23, uL22, and PD-1, is a component of the 60S large ribosomal subunit.
- The protein is essential for protein synthesis, as part of the ribosome, a large ribonucleoprotein complex responsible for translating mRNA into proteins.

PMID:23636399
PMID:32669547 - RPL17 belongs to the universal ribosomal protein uL22 family, conserved across prokaryotes and eukaryotes.
- Subcellularly, RPL17 is localized to the cytoplasm, consistent with its translation machinery role.
- Expression is observed in pancreas, lung, colon, cystic duct, gall bladder, kidney, and liver, with variable levels in pancreatic tumor cell lines; high in well-differentiated lines (e.g., HPAF) and lower in poorly differentiated ones (e.g., PANC-1).
- The protein is subject to alternative splicing, and mutations or dysfunction are associated with Diamond-Blackfan anemia, a ribosomopathy.
Experimental Guidance and Technical Tips
- The immunogen encompasses full-length RPL17 (amino acids 1–184), making the antibody likely to recognize endogenous protein; cross-reactivity with mouse and rat suggests conserved epitopes.
- For western blot, expect a band near 21 kDa; consider loading controls and validate the assay with known expression models.
- In immunohistochemistry, antigen retrieval conditions may need optimization for formalin-fixed paraffin-embedded tissues; pancreas or colon tissues are suitable positive controls.
- For immunofluorescence, fixation and permeabilization should be optimized to maintain ribosomal antigen integrity.
- ELISA can be employed for quantitative analysis; if developing a sandwich immunoassay, consider pairing with a capture antibody targeting a different epitope.
- As RPL17 expression can vary with cell proliferation status, normalize signals against total protein or a stably expressed ribosomal protein like RPLP0.
