Biological Background: RPL11 Function and Localization
- RPL11, also known as 60S ribosomal protein L11 or large ribosomal subunit protein uL5, is a component of the large ribosomal subunit.
- As a core ribosomal protein, RPL11 participates in protein synthesis by contributing to the peptidyl transferase center (PTC) activity, which catalyzes peptide bond formation during translation (PubMed:19191325, PubMed:32669547). Related references: PMID:19191325, PMID:32669547

- Beyond its structural role, RPL11 couples ribosome biogenesis to p53/TP53 activation. Upon ribosomal stress, the 5S RNP accumulates in the nucleoplasm and inhibits MDM2, leading to TP53 stabilization and activation (PubMed:24120868). Related references: PMID:24120868
- RPL11 localizes predominantly to the nucleolus and cytoplasm, and is also found in the nucleoplasm under certain conditions. It promotes nucleolar localization of PML.
- Post-translational modifications include acetylation, phosphorylation, and isopeptide bond formation. The protein is also associated with Diamond-Blackfan anemia when mutated.
