Biological Background: RPL11 Function and Localization
- RPL11 encodes the human 60S ribosomal protein L11, also known as uL5, L11, DBA7, and GIG34. It is a core component of the large ribosomal subunit.
- As part of the ribosome, RPL11 contributes to the peptidyl transferase center (PTC) that catalyzes peptide bond formation during protein synthesis. Related references: PMID:19191325 PMID:32669547
- RPL11 is an essential component of the 5S ribonucleoprotein particle (5S RNP), required for large subunit assembly and rRNA maturation. Related references: PMID:12962325 PMID:19061985 PMID:24120868
- Under nucleolar stress, RPL11 accumulates in the nucleoplasm, where it binds and inhibits MDM2, leading to stabilization and activation of the tumor suppressor p53. Related references: PMID:24120868
- RPL11 localizes to the nucleolus and cytoplasm, dynamically shuttling in response to ribosome biogenesis status.
- The protein undergoes post-translational modifications including acetylation, phosphorylation, and ubiquitination, which may modulate its function and stability.
- Mutations in RPL11 are associated with Diamond-Blackfan anemia, a congenital bone marrow failure syndrome.
- RPL11 promotes the nucleolar localization of PML protein (By similarity).
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 1–177 of human RPL11, representing the N-terminal region. This antibody may detect both full-length and truncated RPL11 species.
- For immunofluorescence (IF/ICC), consider optimizing permeabilization to visualize nucleolar and cytoplasmic RPL11 signals in human cell lines.
- In ELISA applications, empirical determination of coating antigen concentration and antibody dilution is recommended to achieve optimal signal-to-noise ratios.
- Given the high conservation of ribosomal proteins, cross-reactivity with RPL11 orthologs from other species should be validated if intended for non-human samples.
