- Functions as a key effector for the small GTPase RAB3A, playing an essential role in the docking and fusion steps of regulated exocytosis, including neurotransmitter release and hormone secretion.
- At the presynaptic terminal, RPH3A is recruited by GTP-bound RAB3A to the synaptic vesicle membrane, where it modulates synaptic vesicle trafficking and calcium-triggered neurotransmitter release.
- In the postsynaptic compartment, RPH3A forms a ternary complex with GRIN2A (NMDA receptor subunit) and DLG4 (PSD-95), contributing to the stabilization of NMDA receptors at the synapse.
- Localizes to multiple membrane-associated compartments: predominantly found on the cytoplasmic side of synaptic vesicle membranes, but also detected at the postsynaptic cell membrane and within dendritic spines.
