Biological Background: RPA32/RPA2 Function and Localization

  • RPA2 encodes the 32 kDa subunit of the heterotrimeric replication protein A (RPA) complex, also known as replication factor A protein 2 (RF-A p2) or RP-A p34.
  • The RPA complex binds and stabilizes single-stranded DNA (ssDNA) intermediates formed during DNA replication or upon DNA stress, preventing reannealing and recruiting proteins involved in DNA metabolism.
  • In the DNA damage response, RPA2 facilitates repair and checkpoint activation by recruiting ATRIP/ATR kinase; recruits RAD51 and RAD52 for double-strand break repair; recruits XPA and XPG for nucleotide excision repair; and interacts with UNG for base excision repair.
  • RPA stimulates 5'-3' helicase activity of BRIP1/FANCJ. Related references: PMID:17596542
  • The RPA complex also contributes to telomere maintenance.
  • Subcellular localization includes nucleus and PML bodies.
  • Post-translational modifications such as acetylation, phosphorylation, and ubiquitination regulate RPA2 function; alternative splicing generates multiple isoforms.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 1-270, covering nearly the full-length protein; the antibody is likely to recognize native conformations suitable for multiple applications.
  • In Western blotting, expect a band approximately 29 kDa; validate in human, mouse, and rat lysates.
  • For immunofluorescence and immunohistochemistry, anticipate nuclear staining consistent with RPA2 localization; consider appropriate fixation and permeabilization protocols.
  • For ELISA, titrate the antibody to optimize signal-to-noise ratio in your specific assay system.
