Biological Background: Tyrosine-protein kinase transmembrane receptor ROR2 Function and Localization
- ROR2 is a tyrosine-protein kinase receptor also known as neurotrophic tyrosine kinase receptor-related 2 (NTRKR2).
- It is involved in early chondrocyte formation and is required for cartilage and growth plate development (by similarity).
- Phosphorylates YWHAB, leading to induction of osteogenesis and bone formation. Related references: PMID:17717073
- In vitro studies have shown that ROR2 may exhibit very little tyrosine kinase activity.
- Functions as a receptor for WNT5A, potentially inhibiting WNT3A-mediated signaling. Related references: PMID:25029443
- Localized to the cell membrane as a single-pass type I membrane protein.
- Features an immunoglobulin domain, kringle domain, and kinase domain; undergoes glycosylation, phosphorylation, and other post-translational modifications.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 100–200 within the extracellular domain of human ROR2; consider validating antibody specificity using knockout or overexpression models.
- For Western blotting, the predicted molecular weight is ~105 kDa; gel percentage and transfer conditions should be optimized accordingly.
- The antibody cross-reacts with mouse and rat ROR2, enabling studies in rodent models of skeletal development and Wnt signaling.
- In ELISA applications, titrate antibody concentrations and validate with appropriate positive and negative controls to ensure robust signal-to-noise ratios.
