Biological Background: ROM1 Function and Localization
- Rod outer segment membrane protein 1 (ROM1), also known as tetraspanin-23 (Tspan-23), is a transmembrane protein belonging to the tetraspanin family.
- ROM1 plays a critical role in rod outer segment (ROS) morphogenesis and the maintenance of the structural integrity of ROS curved disks, likely in cooperation with peripherin-2 (PRPH2).
- It is involved in the organization of the ROS and maintenance of ROS disk diameter, as well as the preservation of the retina outer nuclear layer.
- The protein localizes to the photoreceptor inner segment membrane and outer segment membrane, specifically enriched in the rim region of ROS disks.
- At the protein level, ROM1 is detected in retina photoreceptors, as shown by immunolocalization studies that highlight its presence in the disk rims. Related references: PMID:1610568, PMID:8504299
- It contains conserved tetraspanin features, including four transmembrane domains and disulfide bonds, and is implicated in cell adhesion and sensory transduction.
- Mutations in ROM1 are associated with retinitis pigmentosa, a degenerative retinal disease affecting vision.

- The immunogen is a recombinant fragment corresponding to amino acids 126–263 of human ROM1, which may aid in specific detection across species where this region is conserved.
- The antibody has been applied in Western blot (WB), immunohistochemistry on paraffin-embedded sections (IHC-P), and ELISA. Consider validating these applications in your model system, especially using retinal tissue or cell lysates.
- For IHC-P, antigen retrieval conditions may need optimization, and endogenous peroxidase blocking is recommended for retinal tissues.
- The predicted molecular weight of ROM1 is approximately 37 kDa, but post-translational modifications or dimerization may result in bands at different apparent sizes in WB.
