Biological Background: ROCK2 Function and Localization

  • Rho-associated protein kinase 2 (ROCK2), also known as ROCK-II or p164 ROCK-2, is a serine/threonine-protein kinase encoded by the ROCK2 gene.
  • It serves as a key regulator of the actin cytoskeleton and cell polarity, mediating smooth muscle contraction, stress fiber formation, focal adhesion dynamics, neurite retraction, and cell motility through phosphorylation of multiple substrates including ADD1, CNN1, EZR, MYL9/MLC2, and VIM.
  • ROCK2 phosphorylates and activates MAPK1/3 and RPS6KA1 during myogenic differentiation, and acts as a negative regulator of VEGF-induced angiogenic endothelial cell activation.
  • It plays an important role in centrosome duplication and contributes to the generation of circadian rhythms in vascular contractility by modulating myosin light chain phosphorylation.
  • Subcellularly, ROCK2 localizes to the cytoplasm, cell membrane, nucleus, cytoskeleton, and centrosome.
  • At the protein level, ROCK2 is specifically expressed in the brain, and its structural features include coiled-coil, zinc-finger, and kinase domains (ATP-binding, magnesium- and metal-binding).

  • The immunogen used corresponds to the C‑terminal region (aa 1109–1388) of human ROCK2, which includes the pleckstrin homology and cysteine‑rich domains. This design is expected to generate antibodies recognizing epitopes in the C‑terminal half of the protein.
  • For Western blotting, a band of approximately 160–161 kDa is anticipated in lysates from human, mouse, or rat tissues/cells. Include appropriate positive controls such as brain lysate (known tissue of expression) and validate the specific band using siRNA knockdown or competing peptide if necessary.
  • In immunohistochemistry (IHC‑P) and immunofluorescence (IF/ICC), consider that ROCK2 exhibits broad subcellular distribution (cytoplasm, membrane, nucleus, and centrosome); optimize fixation and permeabilization conditions and include isotype controls.
  • The antibody is also suitable for ELISA; pair with a capture/detection system validated for your matrix. Because it is a polyclonal antibody, batch‑to‑batch variation should be assessed if switching lots.
