- ROCK1 encodes Rho-associated protein kinase 1, a serine/threonine kinase of approximately 158 kDa, featuring coiled-coil, zinc-finger, and pleckstrin homology domains.
- It is a key regulator of the actin cytoskeleton and cell polarity, mediating smooth muscle contraction, stress fiber and focal adhesion formation, neurite retraction, cell adhesion and motility via phosphorylation of substrates such as LIMK1, LIMK2, MYL9/MLC2, and PPP1R12A.
