Biological Background: ROCK1 Function and Localization
- ROCK1 is a serine/threonine-protein kinase and member of the Rho-associated coiled-coil-containing protein kinase family. It is also known as ROCK-I or p160ROCK, and was identified as a renal carcinoma antigen NY-REN-35.
- ROCK1 is a key regulator of the actin cytoskeleton and cell polarity, controlling smooth muscle contraction, actin organization, stress fiber and focal adhesion formation, neurite retraction, cell adhesion and motility through phosphorylation of substrates like DAPK3, GFAP, LIMK1/2, MYL9/MLC2, PFN1 and PPP1R12A. - The immunogen covers amino acids 400–660 within the N-terminal half of ROCK1, which may allow recognition of full-length protein and potentially some isoforms. Verify reactivity in the relevant sample system.
- For Western blot (WB), the predicted molecular weight is approximately 158 kDa. Use appropriate gel percentage (e.g., 6–8% SDS‑PAGE) and optimize transfer conditions for high‑molecular‑weight proteins.
- For immunoprecipitation (IP), titrate the antibody amount and select a suitable binding/wash buffer to maximize capture efficiency.
- For immunofluorescence on paraffin‑embedded sections (IF‑P), consider heat‑induced antigen retrieval and include both positive and negative control tissues.
