Biological Background: ROCK1 Function and Localization
-
ROCK1 is a serine/threonine-protein kinase that acts as a master regulator of the actin cytoskeleton, driving essential processes such as cell polarity, smooth muscle contraction, stress fiber and focal adhesion formation, neurite retraction, cell adhesion, and motility. These effects are mediated through phosphorylation of diverse substrates including DAPK3, GFAP, LIMK1, LIMK2, MYL9/MLC2, TPPP, PFN1, and PPP1R12A (PubMed:10436159, PubMed:10652353, PubMed:11018042). Phosphorylation of FHOD1 by ROCK1 synergistically promotes SRC-dependent non-apoptotic plasma membrane blebbing (PubMed:18694941). Related references: PMID:18694941
-
Under UVB stress, ROCK1 phosphorylates JIP3 and modulates the consequent recruitment of JNK, controlling downstream stress signaling (PubMed:19036714). Related references: PMID:19036714
-
ROCK1 negatively regulates VEGF-induced angiogenic endothelial cell activation, thereby limiting excessive angiogenesis (PubMed:19181962). Related references: PMID:19181962
-
ROCK1 is involved in terminal differentiation processes, contributing to erythroid differentiation (PubMed:21072057) and promoting keratinocyte terminal differentiation (PubMed:19997641). Related references: PMID:21072057 PMID:19997641
-
Subcellularly, ROCK1 is distributed broadly across the cytoplasm, cytoskeleton (centrosome/centriole), Golgi apparatus membrane, cell membrane, and dynamic cell projections including blebs, lamellipodia, and ruffles. It is detected in blood platelets.
-
Structurally, ROCK1 contains a kinase domain, coiled-coil region, and zinc-finger motifs, and its activity is modulated by phosphorylation and acetylation.
Experimental Guidance and Technical Tips
- For western blotting, note that full-length ROCK1 has a predicted molecular weight of approximately 158 kDa. Select the appropriate polyacrylamide gel percentage (e.g., 6–8%) and optimize transfer conditions for high-molecular-weight proteins. It is advisable to validate band specificity using peptide competition assays or siRNA-mediated knockdown.
- This antibody is confirmed for reactivity with human, mouse, and rat samples; when using lysates from other species, cross-reactivity should be verified experimentally.
- For ELISA applications, it is recommended to pair the antibody with a matched detection antibody and to establish standard assay linearity and sensitivity with recombinant ROCK1 protein or relevant biological samples.
- Given the immunogen design (a peptide within residues 1100–1200), consider that the epitope lies within the C-terminal region; denaturing conditions or partial proteolysis may affect detection efficiency.
