Biological Background: ROBO1 Function and Localization
- ROBO1 (Roundabout homolog 1), also known as DUTT1 (Deleted in U Twenty Twenty) or H-Robo-1, is a receptor for SLIT1 and SLIT2 guidance cues.
- It mediates axonal navigation at the ventral midline of the neural tube and projection of axons during neuronal development. Related references: PMID:10102268, PMID:24560577
- Interaction with the intracellular domain of FLRT3 mediates axon attraction towards cells expressing NTN1. Related references: PMID:24560577
- The protein is localized at the cell membrane, axon projections, and the endoplasmic reticulum-Golgi intermediate compartment membrane.
- It is widely expressed across tissues, with the exception of kidney.
- ROBO1 regulates cell migration by interacting with MYO9B, leading to increased levels of active, GTP-bound RHOA. Related references: PMID:26529257
- The protein contains immunoglobulin domains, fibronectin type III repeats, and a transmembrane domain, and is subject to glycosylation, phosphorylation, and ubiquitination.
- Alternatively spliced isoforms and disease-associated variants (e.g., CPHD8, dwarfism, intellectual disability) highlight its importance in development and disease.
Experimental Guidance and Technical Tips
- For Western blot, anticipate a band around 181 kDa; include molecular weight markers and validate with a known positive control (e.g., HEK293 or mouse brain lysate).
- The immunogen spans amino acids 1452–1651 in the C-terminal region; confirm specificity by pre-incubating the antibody with the immunogen peptide in blocking experiments.
- For IF/ICC, use appropriate cell lines expressing ROBO1 (e.g., neuroblastoma or cortical neuron cultures) and perform permeabilization steps; co-staining with axonal markers can help verify localization.
- Given the cross-reactivity with mouse and rat, this antibody may be suitable for rodent models of neurodevelopment; perform initial titration in the intended species.
- In ELISA, consider using recombinant ROBO1 extracellular domain fragments as capture reagents and validate with SLIT1/2 binding competition assays.
