Biological Background: RNF8 Function and Localization
- RNF8 is an E3 ubiquitin-protein ligase that plays a central role in DNA damage signaling by mediating the 'Lys-63'-linked ubiquitination of histones H2A and H2AX at double-strand breaks (DSBs), thereby promoting the recruitment of DNA repair proteins such as TP53BP1 and BRCA1 (PubMed:18001824, 18006705). - It also catalyzes 'Lys-48'-linked polyubiquitin chains, targeting substrates like JMJD2A/KDM4A for degradation, which unmasks H4K20me2 marks and facilitates TP53BP1 recruitment at DNA damage sites (PubMed:11322894, 14981089, 17724460).
