Biological Background: RNF7 Function and Localization

  • RNF7 (also known as ROC2, SAG, or CKBBP1) is the catalytic component of cullin-5-RING E3 ubiquitin-protein ligase complexes (ECS complexes), which mediate the ubiquitination and subsequent proteasomal degradation of target proteins.   • The functional specificity of ECS complexes is determined by the SOCS box-containing substrate recognition subunit. For example, RNF7-containing ECS(SOCS2) complex ubiquitinates phosphorylated EPOR and GHR, leading to their degradation. Related references: PMID:21980433, PMID:25505247
  • RNF7 specifically catalyzes the neddylation of cullin-5 (CUL5) via interaction with the E2 enzyme UBE2F, but does not neddylate other cullins (CUL1, CUL2, CUL3, CUL4A, or CUL4B). Related references: PMID:19250909
  • RNF7 localizes to both the cytoplasm and nucleus, consistent with its roles in degrading cytosolic and nuclear substrates.
