Biological Background: RNF7 Function and Localization
- RNF7 (also known as ROC2, SAG, or CKBBP1) is the catalytic component of cullin-5-RING E3 ubiquitin-protein ligase complexes (ECS complexes), which mediate the ubiquitination and subsequent proteasomal degradation of target proteins. Related references: PMID:21980433, PMID:33268465, PMID:38418882
- The functional specificity of ECS complexes is determined by the SOCS box-containing substrate recognition subunit. For example, RNF7-containing ECS(SOCS2) complex ubiquitinates phosphorylated EPOR and GHR, leading to their degradation. Related references: PMID:21980433, PMID:25505247
- RNF7 specifically catalyzes the neddylation of cullin-5 (CUL5) via interaction with the E2 enzyme UBE2F, but does not neddylate other cullins (CUL1, CUL2, CUL3, CUL4A, or CUL4B). Related references: PMID:19250909
- RNF7 localizes to both the cytoplasm and nucleus, consistent with its roles in degrading cytosolic and nuclear substrates.
- In humans, RNF7 is expressed primarily in heart, liver, skeletal muscle, and pancreas, with very low expression in brain, placenta, and lung.
- RNF7 may play a role in protecting cells from apoptosis induced by redox agents, and it is hijacked by HIV-1 Vif to degrade host antiviral factors APOBEC3F and APOBEC3G.
Experimental Guidance and Technical Tips
- The immunogen for this antibody is a recombinant fusion protein corresponding to amino acids 1-113 of human RNF7 (NP_055060.1). This region covers the N-terminal portion including the RING finger domain, critical for its E3 ligase activity.
- For Western blot (WB), consider that endogenous RNF7 is approximately 13 kDa. Ensure appropriate gel percentage and transfer conditions for small proteins.
- In immunofluorescence (IF/ICC), validate fixation and permeabilization protocols, as RNF7 is present in both cytoplasm and nucleus.
- When using ELISA, optimize coating antigen and antibody dilutions for your specific assay format.
