Biological Background: RNF7 Function and Localization

  • RNF7 (SAG, ROC2, RBX2, CKBBP1) is a RING-box protein and the catalytic component of multiple cullin-5-RING E3 ubiquitin-protein ligase (ECS) complexes, which mediate ubiquitination and proteasomal degradation of target proteins, thereby regulating cell cycle progression, signal transduction, and transcription.

  • Within various SOCS box-containing ECS complexes, RNF7 targets a broad range of substrates for degradation, including EPOR, GHR, CKB, CGAS, ANKRD28, SUV39H1, BTRC, and CDO1, via specific adaptors such as SOCS2, ASB9, SPSB3, RAB40C, ASB7, and LRRC58 (PubMed:21980433, PubMed:25505247, PubMed:33268465, PubMed:38418882, PubMed:35512830, PubMed:40440427, PubMed:27910872, PubMed:40963025).

    PMID:40963025

  • RNF7 specifically catalyzes the neddylation of cullin-5 (CUL5) through interaction with UBE2F, and does not neddylate other cullins (PubMed:19250909).

    PMID:19250909

  • Subcellular localization includes both the cytoplasm and nucleus.

  • Expressed predominantly in heart, liver, skeletal muscle, and pancreas, with very low levels in brain, placenta, and lung.

  • Post-translational modifications include acetylation and phosphorylation; the protein contains a zinc-finger RING-type domain essential for E3 ligase activity, and is classified as a transferase in the ubiquitin conjugation pathway.

  • Plays a role in protecting cells from apoptosis induced by redox agents (PubMed:10082581).

  • During HIV-1 infection, RNF7 is hijacked by the viral Vif protein to form an ECS complex that targets host APOBEC3F and APOBEC3G for degradation, counteracting innate antiviral defenses.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1–113) of human RNF7. This region is likely to be well-conserved across species, consistent with the observed cross-reactivity to mouse and rat.
  • For Western blotting (WB), consider a starting dilution range of 1:500–1:2000 and use a molecular weight marker that includes the ~13 kDa band expected for RNF7.
  • In immunohistochemistry (IHC-P), validate the antibody using positive and negative control tissues and optimize antigen retrieval conditions. Perform additional validation in mouse or rat tissues if needed.
  • For ELISA applications, the recombinant immunogen can serve as a positive control to assess antibody binding.
  • Given the involvement of RNF7 in ubiquitination pathways, consider using appropriate inhibitors (e.g., proteasome inhibitors) to stabilize target proteins in functional assays if required.
  • Always validate the antibody’s performance in your specific experimental system and sample type.
