Biological Background: RNF40 Function and Localization
- RNF40 (also known as BRE1B, RBP95, STARING) is the E3 ubiquitin-protein ligase BRE1B, a component of the RNF20/40 complex that catalyzes monoubiquitination of histone H2B at Lys-120 (H2BK120ub1).
- H2BK120ub1 is a key epigenetic mark for transcriptional activation and is required for subsequent methylation of histone H3 at Lys-4 and Lys-79 (H3K4me and H3K79me), linking it to gene regulation.
- The RNF20/40 complex functions in cooperation with the E2 enzymes UBE2A or UBE2B, and is essential for the activation of Hox genes.
- RNF40 localizes to the nucleus, consistent with its role in chromatin modification.
- Ubiquitously expressed, with higher levels in testis, heart, and pancreas, and lower in lung, skeletal muscle, and small intestine.
- Contains a RING-type zinc finger domain and participates in host-virus interactions, promoting the lytic cycle of human herpesvirus 8 (KSHV) by inducing viral gene expression.
- Post-translational modifications include acetylation, phosphorylation, and isopeptide bond formation (ubiquitination-related).
