  • RNF40 encodes the E3 ubiquitin-protein ligase BRE1B, also known as 95 kDa retinoblastoma-associated protein (RBP95) and RING finger protein 40. It is a member of the RING-type E3 ubiquitin transferase family, characterized by a RING finger domain that mediates zinc binding and ubiquitin ligase activity.
  • BRE1B forms a functional complex with RNF20 to catalyze monoubiquitination of histone H2B at lysine 120 (H2BK120ub1). This modification serves as a specific epigenetic mark for transcriptional activation and is essential for subsequent methylation of histone H3 at lysine 4 and lysine 79 (H3K4me and H3K79me).
  • Through its role in H2B ubiquitination, the RNF20/40 complex is central to the histone code and gene regulation. It is required for the transcriptional activation of Hox genes and likely many other developmental and proliferation-related loci.
  • BRE1B is localized predominantly in the nucleus, consistent with its function in chromatin modification and transcriptional control.
  • The protein is ubiquitously expressed in human tissues, with higher levels observed in testis, heart, and pancreas, and lower levels in lung, skeletal muscle, and small intestine.
