Biological Background: RNF4 Function and Localization

• RNF4 (SLX5, SNURF) is an E3 ubiquitin-protein ligase that specifically binds polysumoylated chains and mediates polyubiquitination (Lys-6, Lys-11, Lys-48, Lys-63 linkages), targeting substrates for proteasomal degradation.

• For Western blotting, a band around 21 kDa is expected for unmodified RNF4. Due to extensive post-translational modifications (sumoylation, ubiquitination), additional bands may be observed. Validate in samples with high endogenous RNF4, such as testis tissue.
• In ELISA applications, consider coating with the recombinant immunogen or sumoylated substrates to prove specificity; indirect ELISA conditions should be empirically determined.
• Cross-reactivity with mouse and rat is reported; verify sequence homology in the immunogen region, as the N-terminal sequence is highly conserved among these species.
