Biological Background: RNF34 Function and Localization
- RNF34 is an E3 ubiquitin-protein ligase that catalyzes the ubiquitin-mediated proteasomal degradation of target proteins, including CASP8 and CASP10, thereby negatively regulating extrinsic apoptosis. Related references: PMID:15069192

Consider validating the antibody’s specificity in your target cell lines using appropriate controls.
- For Western blot (WB), the predicted molecular weight of RNF34 is approximately 42 kDa. A band near this size can be expected, but post-translational modifications or isoform variability may lead to slight shifts. Optimize blocking and antibody dilution for your samples.
- In immunofluorescence (IF/ICC), RNF34 has been reported to localize to the nucleus (including nuclear speckles), cytosol, and cell membrane. Proper fixation and permeabilization conditions should be tested to preserve antigenicity and achieve specific staining.
- For ELISA, the polyclonal nature of the antibody may provide high sensitivity for detecting RNF34 in human samples. Include standard curves and negative controls to ensure assay linearity and specificity.
