Biological Background: RNF220 Function and Localization

  • RNF220 is a RING-type E3 ubiquitin-protein ligase that mediates ubiquitination of target proteins, playing roles in Wnt signaling, neurodevelopment, and immune responses.
  • It ubiquitinates and promotes proteasomal degradation of AMPA receptor subunits GRIA1 and GRIA2, thereby modulating synaptic activity and brain function. Related references: PMID:25266658, PMID:33964137
  • Together with ZC4H2, RNF220 monoubiquitinates PHOX2A and PHOX2B, enhancing their DNA-binding ability and transactivity (By similarity).
  • RNF220 stabilizes CTNNB1 (β-catenin) through USP7-mediated deubiquitination, thereby promoting Wnt signaling. Related references: PMID:25266658, PMID:33964137
  • It regulates type I and type II interferon responses by mediating 'Lys-63'-linked polyubiquitination of STAT1, and also regulates STAT3 phosphorylation and transcriptional activity via ubiquitination. Related references: PMID:32814877, PMID:41219491
  • RNF220 promotes 'Lys-63'-linked polyubiquitination and stabilization of OLIG1 and OLIG2, transcription factors essential for oligodendroglial development. Related references: PMID:38324685
  • Subcellular localization: Found in both the cytoplasm and nucleus, consistent with its roles in nuclear signaling and cytoplasmic protein degradation.
  • Tissue specificity: Ubiquitously expressed, with abundance in brain and spinal cord, particularly cerebellum and cerebral cortex; also highly expressed in fetal cerebellum, spinal cord, and cortex.

Experimental Guidance and Technical Tips

  • For Western blotting, expect a band at ~63kDa; validate in human, mouse, or rat lysates. Consider initial dilutions of 1:500-1:2000 with appropriate controls.
  • For ELISA, the polyclonal nature allows use as capture or detection antibody; optimize coating concentrations and assay conditions.
  • Because the immunogen spans amino acids 267-566, epitopes may be conserved across species (cross-reactivity confirmed for human, mouse, rat), but always verify reactivity in your specific sample system.
  • Include positive and negative controls, and perform standard blocking and washing steps to minimize background.
