Biological Background: RNF166 Function and Localization
- RNF166 is an E3 ubiquitin-protein ligase containing a RING finger domain, also known as RING finger protein 166 or RING-type E3 ubiquitin transferase RNF166.
- It promotes the ubiquitination of substrates such as TRAF3, TRAF6, and SQSTM1, participating in diverse biological processes including interferon production and xenophagy.
- Plays a critical role in RNA virus-induced interferon-beta production by mediating the ubiquitination of TRAF3 and TRAF6, activating innate immune responses. Related references: PMID:26456228
- Facilitates the early recruitment of autophagy adapters to bacteria and mediates Lys-29 and Lys-33-linked ubiquitination of SQSTM1, leading to xenophagic targeting and inhibition of bacterial replication. Related references: PMID:27880896
- RNF166 localizes predominantly to the cytoplasm, where it exerts its ubiquitin ligase activity.

Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering amino acids 1–237, which may allow recognition of multiple isoforms if the region is conserved. Validate specificity in your sample system.
- For western blotting, expected band size is approximately 26 kDa; consider using appropriate positive and negative controls.
- For immunofluorescence/ICC, optimize fixation and permeabilization conditions, as the target is cytoplasmic. Counterstaining for nuclei may help confirm localization.
