Biological Background: RNF166 Function and Localization
- RNF166 is an E3 ubiquitin-protein ligase that contains a RING finger domain and zinc-binding sites, characteristic of the RING finger family of ubiquitin transferases.
- It mediates both 'Lys-29' and 'Lys-33'-linked polyubiquitination, distinct from the more common 'Lys-48' and 'Lys-63' linkages, targeting substrates for non-degradative signaling or autophagy. Related references: PMID:27880896
- RNF166 promotes RNA virus-induced interferon-beta production by catalyzing the ubiquitination of TRAF3 and TRAF6, key adaptors in innate immune pathways. Related references: PMID:26456228
- It plays a critical role in xenophagy, the selective autophagy of intracellular bacteria, by ubiquitinating the autophagy receptor SQSTM1/p62 and recruiting autophagy adapters. Related references: PMID:27880896
- RNF166 is predominantly localized in the cytoplasm, positioning it at the hub of immune signaling and intracellular quality control.
- The gene undergoes alternative splicing, generating multiple isoforms that may expand its functional repertoire.
- As a RING-type E3 ligase, RNF166 directly transfers ubiquitin from E2 enzymes to specific substrates, thereby determining the cellular fate of target proteins.
Experimental Guidance and Technical Tips
- For Western blotting, the expected molecular weight of RNF166 is approximately 26 kDa. Include a positive control lysate from human, mouse, or rat cell lines known to express RNF166.
- In IHC-P, consider optimizing antigen retrieval conditions and primary antibody dilution on known positive tissue sections, such as human spleen or lymph node, before proceeding to experimental samples.
- For ELISA, the immunogen peptide can serve as a positive control. Given cross-reactivity with mouse and rat, the antibody may be used to quantify RNF166 across these species after validation.
