Biological Background: RNF144A Function and Localization
- RNF144A, also known as UIP4, RNF144, or UBCE7IP4, is an E3 ubiquitin-protein ligase containing a RING finger domain and a transmembrane helix.
- It accepts ubiquitin from E2 enzymes UBE2L3 and UBE2L6 and directly transfers it to substrate proteins, functioning as a key component of the ubiquitination machinery. Related references: PMID:26216882
- During DNA damage, RNF144A mediates the ubiquitination and degradation of the DNA damage kinase PRKDC, contributing to DNA repair regulation. Related references: PMID:24979766
- It positively regulates innate immune responses by mediating STING1 ubiquitination and promoting its translocation from the endoplasmic reticulum to the Golgi, thereby activating downstream signaling pathways. Related references: PMID:37955227
- RNF144A plays a positive role in EGF-dependent cell proliferation by prolonging EGFR signaling through EGFR ubiquitination. Related references: PMID:30171075
- Independently of EGFR, it increases ERK activity by promoting polyubiquitination and subsequent degradation of VRK3 in the cytosol. Related references: PMID:33067254
- Subcellularly, RNF144A is found at the cell membrane, cytoplasmic vesicle membrane, endosome membrane, and endoplasmic reticulum membrane.
- The protein features a zinc-finger RING-type domain essential for its ligase activity and a transmembrane region anchoring it to membranes.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 52-179 of human RNF144A, targeting the N-terminal region; this region is present in the full-length protein and may detect isoforms.
- For Western blot, a band around 33 kDa is expected in human and rat lysates; validate molecular weight in your specific sample system and consider using loading controls.
- In ELISA applications, optimize coating antigen concentration and antibody dilution; the polyclonal nature may provide broad epitope recognition suitable for sandwich assay development.
- Confirm specificity by using RNF144A knockout or knockdown cell lines, as the antibody may cross-react with related RING finger proteins.
- For immunohistochemistry or other untested applications, pilot experiments are recommended to determine suitability.
