Biological Background: RNF14 Function and Localization
- Also known as Androgen receptor-associated protein 54 (ARA54), HFB30, and RING finger protein 14. Multiple isoforms exist due to alternative splicing.
- Functions as an E3 ubiquitin-protein ligase in the RNF14-RNF25 translation quality control pathway, mediating 'Lys-6'-linked ubiquitination of translation factors (EEF1A1, ETF1) and ribosomal proteins on stalled ribosomes (PubMed:36638793, PubMed:37651229, PubMed:37951215). Related references: PMID:36638793, PMID:37651229, PMID:37951215
- Required for resolving RNA-protein cross-links caused by reactive aldehydes, promoting degradation of cross-linked products via the VCP/proteasome pathway (PubMed:37951215, PubMed:37951216). Related references: PMID:37951215, PMID:37951216
- Acts as a transcriptional regulator in Wnt signaling through interaction with TCF transcription factors TCF7/TCF1, TCF7L1/TCF3, and TCF7L2/TCF4 (PubMed:23449499). Related references: PMID:23449499
- May function as a coactivator for androgen- and progesterone-dependent transcription (PubMed:19345326). Related references: PMID:19345326
- Subcellular localization includes both cytoplasm and nucleus.
- Widely expressed across tissues.
- Contains zinc-finger domains and is subject to phosphorylation; classified under transferase, Ubl conjugation pathway, and metal-binding proteins.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the internal region (aa 300–400) of human RNF14; this may favor recognition of denatured linear epitopes, making the antibody well-suited for Western blotting (WB) under reducing/denaturing conditions.
- For ELISA applications, consider using the immunizing peptide as a positive coating antigen and validate the antibody's performance with full-length recombinant protein or cell lysates from human, mouse, or rat sources to confirm cross-reactivity.
- Since RNF14 is expressed in both cytoplasm and nucleus, ensure appropriate fractionation or total lysate preparation to detect the target in WB.
- Verify antibody specificity by testing in RNF14-overexpressing or knockdown cell lines, if available.
- Use appropriate loading controls (~54 kDa) and confirm the observed band size matches the predicted molecular weight of RNF14.
