Biological Background: RNF126 Function and Localization
- RNF126 (RING finger protein 126) is an E3 ubiquitin-protein ligase featuring a RING-type zinc finger domain.
- Mediates 'Lys-27'-, 'Lys-29'-, 'Lys-48'- and/or 'Lys-63'-linked polyubiquitination of target proteins, with the linkage type dependent on the associated E2 enzyme. Related references: PMID:36563124

Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 200–300 of human RNF126 (NP_919442.1). This region was selected for specificity; cross-reactivity with other proteins should be ruled out by appropriate controls.
- For Western blot, expect a major band at approximately 34 kDa. Additional bands may appear due to ubiquitination or other post-translational modifications; include positive and negative control lysates.
- In IHC-P and IF/ICC, antigen retrieval and optimized fixation/permeabilization may improve signal detection.
- For ELISA, the antibody can serve as capture or detection reagent after proper pairing validation. Titration is recommended for each assay format.
