Biological Background: RNF11 Function and Localization

• RNF11, also known as CGI-123 or SID1669, is a RING finger protein that acts as an essential component of a ubiquitin-editing complex, which includes TNFAIP3, ITCH, and TAX1BP1, ensuring transient inflammatory signaling.
• It promotes the association of TNFAIP3 with RIPK1 upon TNF stimulation, leading to deubiquitination of Lys-63-linked chains and addition of Lys-48-linked polyubiquitin chains on RIPK1, targeting it for proteasomal degradation and terminating NF-kappa-B activation.
• RNF11 recruits the deubiquitinase STAMBP to the E3 ubiquitin ligase SMURF2, facilitating STAMBP ubiquitination and subsequent degradation by the 26S proteasome.
