Biological Background: RNF10 Function and Localization
- RNF10 (RING finger protein 10, also known as RIE2) is an E3 ubiquitin-protein ligase that plays a critical role in ribosome quality control.
- It catalyzes monoubiquitination of 40S ribosomal proteins RPS2 and RPS3 in response to ribosome stalling during translation initiation (iRQC) or elongation. Related references: PMID:34348161, PMID:34469731, PMID:39609413
- RNF10 activity is triggered by ribosome subunit imbalance, amino acid starvation, or downstream of the EIF2AK4/GCN2-mediated integrated stress response (ISR). Related references: PMID:39609413, PMID:39947182, PMID:39947183
- Monoubiquitinated RPS2 and RPS3 are recognized by RIOK3 kinase, leading to 18S non-functional rRNA decay and degradation of the 40S ribosomal subunit. Related references: PMID:34348161, PMID:34469731, PMID:39609413
- The action of RNF10 in iRQC is counteracted by the deubiquitinase USP10. Related references: PMID:34469731
- RNF10 localizes to both the cytoplasm and nucleus.
- It contains a RING-type zinc finger domain and metal-binding motifs, characteristic of E3 ubiquitin ligases.
- Additional features include alternative splicing, phosphorylation, and involvement in the ubiquitin-conjugation pathway (Keywords: Alternative splicing, Cytoplasm, Metal-binding, Nucleus, Phosphoprotein, Proteomics identification, Reference proteome, Transferase, Ubl conjugation pathway, Zinc, Zinc-finger).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 660–760 of human RNF10 (NP_055683.3), which may confer specificity across human, mouse, and rat targets.
- For Western blotting (WB), the predicted molecular weight is approximately 90 kDa; consider running appropriate controls and validating band sizes in your sample system.
- In immunohistochemistry (IHC-P) and immunocytochemistry (IF/ICC), antigen retrieval and fixation conditions should be optimized, and note the cytoplasmic/nuclear localization of RNF10.
- For ELISA applications, the antibody may be used for capture or detection; validate linearity and sensitivity in your assay buffer.
- Cross-reactivity with other RING finger proteins is unlikely but should be assessed if non-specific signals are observed.
