Biological Background: RMND5A Function and Localization

  • RMND5A is an E3 ubiquitin-protein transferase, also known as p44CTLH, Protein RMD5 homolog A, and GID2.
  • It is a core component of the CTLH E3 ubiquitin-protein ligase complex, which selectively accepts ubiquitin from UBE2H and mediates ubiquitination and proteasomal degradation of the transcription factor HBP1.
  • Both RMND5A and MAEA are required for the catalytic activity of the CTLH E3 ubiquitin-protein ligase complex, and this activity is necessary for normal cell proliferation. Related references: PMID:29911972
  • The CTLH complex is not required for the degradation of enzymes involved in gluconeogenesis, such as FBP1.
  • RMND5A localizes to the nucleus (nucleoplasm) and cytoplasm.
  • It contains a zinc-finger domain and is involved in metal binding, as indicated by UniProt keywords.
  • Post-translational modifications include acetylation; alternative splicing produces multiple isoforms.
