Biological Background: RIPK4 Function and Localization
- Encoded by the RIPK4 gene (also known as ANKRD3, DIK), Receptor-interacting serine/threonine-protein kinase 4 is a member of the RIP kinase family, involved in signal transduction.
- Functions as a serine/threonine protein kinase required for embryonic skin development and adult skin homeostasis; it phosphorylates PKP1 to promote keratinocyte differentiation and cell adhesion (By similarity).
- Acts as a direct transcriptional target of TP63, linking epithelial differentiation to signaling cascades. Related references: PMID:22197488
- Plays a role in NF-kappa-B activation, contributing to inflammatory and immune responses. Related references: PMID:12446564
- Localizes to the cytoplasm and membrane, consistent with its role in membrane-proximal signaling.
- Expressed in hair follicles and skin, aligning with its critical function in epidermal biology.
- Contains ankyrin repeat domains and a kinase domain, and is subject to post-translational modifications, including phosphorylation and ubiquitination (see Keywords: Alternative splicing, ANK repeat, ATP-binding, Isopeptide bond, Phosphoprotein, Ubl conjugation).
- Mutations in RIPK4 are associated with ectodermal dysplasia and related disorders, highlighting its clinical significance (Disease variant keyword).
Experimental Guidance and Technical Tips
- The immunogen was designed against amino acids 240-520 of human RIPK4, which covers the kinase domain and part of the ankyrin repeat region, ensuring reactivity with multiple isoforms.
- For Western blotting, a band at approximately 92 kDa is expected. Use appropriate controls and consider the observed molecular weight may differ slightly due to post-translational modifications.
- IHC-P application may require antigen retrieval; validate in positive control tissues such as skin or hair follicle-containing sections, given the expression pattern.
- Cross-reactivity with mouse makes this antibody suitable for translational studies in murine models of skin development and inflammation.
- For ELISA, consider paired antibody formats for quantitative detection; optimal dilution factors should be determined experimentally.
