Biological Background: RIPK3 (Receptor-interacting serine/threonine-protein kinase 3) Function and Localization

  • Receptor-interacting serine/threonine-protein kinase 3 (RIPK3), also known as RIP3, is a key mediator of programmed necrosis (necroptosis) and apoptosis, functioning downstream of death receptors and pathogen sensors. Related references: PMID:19524512 PMID:19524513 PMID:22265413
  • Upon activation by ZBP1 or TNF receptor signaling, RIPK3 forms a necrosis-inducing complex and phosphorylates MLKL, triggering its translocation to the plasma membrane, calcium influx, and membrane rupture. Related references: PMID:19524512 PMID:19524513 PMID:22265413
  • RIPK3 also regulates apoptosis through a RIPK1-, FADD-, and CASP8-dependent pathway that is independent of MLKL and its own kinase activity.

  • For Western blotting, the predicted molecular weight is approximately 57 kDa; however, additional bands may appear due to post-translational modifications (e.g., phosphorylation) or alternative splicing. Validate band specificity using appropriate controls.
  • For immunohistochemistry (IHC-P), antigen retrieval methods may be required. Positive control tissues include pancreas or colon/lung tumor samples where RIPK3 is highly expressed.
