Biological Background: RING1B/RNF2 Function and Localization

  • Catalyzes monoubiquitination of histone H2A at Lys-119 (H2AK119Ub), a critical epigenetic mark for Polycomb-mediated transcriptional repression. Related references: PMID:15386022, PMID:16359901, PMID:21772249
  • Core component of Polycomb repressive complex 1 (PRC1), essential for maintaining the silenced state of Hox genes and other developmental regulators. Related references: PMID:16359901, PMID:26151332
  • Functions as the principal E3 ubiquitin ligase for H2A within the PRC1 complex; its activity is modulated by the accessory protein RING1.
  • E3 ligase activity is significantly enhanced by the PRC1 subunit BMI1/PCGF4. Related references: PMID:21772249
  • Involved in X chromosome inactivation, participating in the initiation of both imprinted and random inactivation in female mammals.
  • Exhibits cell-cycle-dependent association with chromatin; in resting B- and T-lymphocytes, interaction with AURKB inhibits its activity to maintain basal transcription.

Experimental Guidance and Technical Tips

  • For Western blotting, prepare whole-cell or nuclear extracts under reducing conditions and expect a band near 38 kDa. Use appropriate positive and negative control lysates to confirm specificity.
  • In immunohistochemistry (IHC-P), antigen retrieval conditions and antibody concentration should be optimized for each tissue type. Consider using a biotin-free detection system to minimize background.
  • For immunoprecipitation, mild lysis buffers preserving protein–protein interactions are recommended. Validate co-immunoprecipitated partners by Western blot.
  • In ELISA, the antibody can be used as a capture or detection reagent; perform checkerboard titrations to determine optimal working concentrations.
  • Given the high conservation, cross-reactivity with mouse and rat samples is expected but should be validated in your specific experimental system.
