Biological Background: RING1B/RNF2 Function and Localization
- RING1B/RNF2 is an E3 ubiquitin-protein ligase that mediates monoubiquitination of histone H2A at 'Lys-119' (H2AK119Ub), a critical epigenetic mark for transcriptional repression. Related references: PMID:15386022, PMID:16359901, PMID:21772249
- It is an essential component of the Polycomb repressive complex 1 (PRC1), which maintains the silenced state of developmental genes such as Hox genes through chromatin remodeling and histone modification. Related references: PMID:16359901, PMID:26151332
- The E3 ubiquitin ligase activity of RNF2 is enhanced by its interaction with BMI1/PCGF4. Related references: PMID:21772249
- RNF2 also plays a role in X chromosome inactivation and acts as a negative regulator of autophagy by targeting AMBRA1 for degradation.
- Subcellular localization includes the nucleus, cytoplasm, and chromosomes, and its association with chromatin is cell-cycle dependent.
- The protein contains a zinc-finger domain and is subject to post-translational modifications such as acetylation and phosphorylation; mutations are linked to epilepsy and intellectual disability.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human RING1B/RNF2 protein (amino acids 1–336). For applications like Western blotting (WB) and immunoprecipitation (IP), consider validating the antibody in relevant cell lines expressing RNF2.
- Given its involvement in chromatin biology, this antibody may be suitable for chromatin immunoprecipitation (ChIP) experiments to study histone modifications or PRC1 complex binding. Validate enrichment under appropriate fixation conditions.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), test antigen retrieval methods and antibody dilutions in control tissues or cells with known expression patterns.
