Biological Background: Rig-I/DDX58 Function and Localization

  • RIG-I is a cytoplasmic innate immune receptor that recognizes viral double-stranded RNA and 5'-triphosphorylated single-stranded RNA, triggering a signaling cascade that leads to type I interferon and pro-inflammatory cytokine production.   • Upon RNA binding, RIG-I homooligomerizes into filaments and recruits the E3 ubiquitin ligase RNF135, which activates and amplifies antiviral signaling through ubiquitination-dependent and -independent mechanisms. Related references: PMID:28469175, PMID:31006531
