Biological Background: Rig-I/DDX58 Function and Localization
- RIG-I is a cytoplasmic innate immune receptor that recognizes viral double-stranded RNA and 5'-triphosphorylated single-stranded RNA, triggering a signaling cascade that leads to type I interferon and pro-inflammatory cytokine production. - Upon RNA binding, RIG-I homooligomerizes into filaments and recruits the E3 ubiquitin ligase RNF135, which activates and amplifies antiviral signaling through ubiquitination-dependent and -independent mechanisms. Related references: PMID:28469175, PMID:31006531
- RIG-I detects a broad spectrum of RNA viruses, including influenza, measles, hepatitis C, dengue, and West Nile viruses, as well as SARS-CoV-2; it also senses some DNA viruses like Epstein-Barr virus via RNA polymerase III-transcribed RNAs. - For Western blot, the predicted molecular weight is approximately 107 kDa. Use appropriate protein markers and consider loading controls to confirm band size.
- The immunogen corresponds to amino acids 241–455, spanning the helicase core domain. This region is conserved across human, mouse, and rat, consistent with the reported cross-reactivity; however, validation in target species is recommended.
- For ELISA, pair with a detection antibody targeting a non-overlapping epitope to avoid competition. Optimize coating and blocking conditions for best signal-to-noise ratio.
