Biological Background: Rig-I/DDX58 Function and Localization

  • RIG-I (retinoic acid-inducible gene I), also known as DDX58, is an ATP-dependent RNA helicase belonging to the RIG-I-like receptor (RLR) family of innate immune sensors.
  • Functions as a cytoplasmic innate immune receptor that detects viral nucleic acids and triggers signaling cascades leading to type I interferon and pro-inflammatory cytokine production. Related references: PMID:15208624, PMID:15708988, PMID:16125763

Experimental Guidance and Technical Tips

  • For Western blotting, the predicted molecular weight is approximately 107 kDa. Consider using positive controls such as virally infected human cell lines (e.g., A549 stimulated with poly(I:C) or infected with Sendai virus) to verify RIG-I induction, as basal expression may be low in unstimulated cells.
  • Since the immunogen is a synthetic peptide, the antibody is expected to recognize a linear epitope. Validate detection of endogenous RIG-I in your sample system and optimize blocking and washing conditions to reduce nonspecific binding.
  • In ELISA, the antibody can be employed as a capture or detection reagent. For quantitative assays, pairing with a matched detection antibody is recommended.
