Biological Background: RICTOR Function and Localization

  • RICTOR (Rapamycin-insensitive companion of mTOR), also known as hAVO3, is a component of the mTORC2 complex, which transduces signals from growth factors to pathways involved in proliferation, cytoskeletal organization, lipogenesis and anabolic output. Related references: PMID:15268862, PMID:15718470, PMID:19720745
  • mTORC2 phosphorylates and activates AGC kinases including AKT (at Ser473), PKC, and SGK1, and is nutrient-insensitive in contrast to mTORC1. Related references: PMID:19720745, PMID:19935711, PMID:19995915, PMID:15467718, PMID:21343617
  • Within mTORC2, RICTOR acts as a molecular adapter that confers rapamycin insensitivity by blocking FKBP12-rapamycin binding. Related references: PMID:21343617, PMID:33158864, PMID:35926713
  • Subcellularly, RICTOR localizes to the cell membrane, endoplasmic reticulum membrane, and lysosome membrane, enabling spatially restricted signaling.
  • RICTOR undergoes multiple post-translational modifications including acetylation, phosphorylation, and ubiquitination (isopeptide bond), as identified by proteomics.
  • Additionally, RICTOR is involved in host-virus interactions and developmental processes, consistent with its classification as a developmental protein.

Experimental Guidance and Technical Tips

  • Given the immunogen design (aa 1043-1142), this antibody may detect full-length RICTOR (~192 kDa) in denaturing western blots; confirm specificity with knockout controls.
  • For ELISA applications, consider pairing with an antibody against a non-overlapping epitope to enhance sensitivity.
  • Optimize blocking and antibody dilution conditions for your specific sample type, as endogenous RICTOR expression levels vary across tissues.
