Biological Background: RHOT2 Function and Localization
- RHOT2, also known as MIRO-2 or ARHT2, is a mitochondrial Rho GTPase and member of the Ras superfamily, sharing structural similarity with RHOT1.
- It serves as an atypical mitochondrial NTPase that hydrolyzes GTP, ATP, and UTP to regulate mitochondrial trafficking and subcellular distribution along microtubules. Related references: PMID:16630562, PMID:22396657, PMID:30513825
- RHOT2 is likely involved in anterograde mitochondrial transport, ensuring proper mitochondrial positioning within cells. Related references: PMID:22396657
- It localizes to the mitochondrial outer membrane, where it functions as an adaptor linking mitochondria to motor proteins.
- The protein is ubiquitously expressed, with highest levels in heart, liver, skeletal muscle, kidney, and pancreas.
- RHOT2 features conserved GTPase and calcium-binding EF-hand domains, and is subject to alternative splicing and ubiquitination (isopeptide bond formation).
- Its predicted molecular mass is approximately 68 kDa (UniProt Q8IXI1), consistent with the observed band in western blot.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 340–550 of human RHOT2) lies within a region that may be conserved across species; cross-reactivity with human, mouse, and rat has been observed. For other species, perform sequence alignment before use.
- For WB, a band at ~68 kDa is expected; ensure adequate membrane protein extraction given mitochondrial localization.
- For IHC-P and IF/ICC, a typical mitochondrial staining pattern should be observed; include appropriate organelle markers to confirm specificity.
- For ELISA, consider pairing with a matched detection antibody and validate in the relevant sample matrix.
