Biological Background: RhoA Function and Localization
- Small GTPase of the Rho family, encoded by the RHOA gene (aliases: ARHA, ARH12, RHO12). Cycles between active GTP-bound and inactive GDP-bound states.
- Key regulator of actin cytoskeleton organization, focal adhesion assembly, and stress fiber formation, linking plasma membrane receptors to cytoskeletal dynamics. Related references: PMID:8910519 PMID:9121475
- Mediates cell migration, adhesion turnover, and cell cycle progression, including cytokinesis and cleavage furrow formation. Related references: PMID:12900402 PMID:16236794 PMID:19934221 PMID:20974804 PMID:23940119 PMID:23871831
- Participates in microtubule-dependent signaling: stabilizes microtubules at the cell cortex via the MEMO1-RHOA-DIAPH1 pathway and regulates APC/CLASP2 localization. Related references: PMID:20937854
- Modulates ion channel activity by promoting KCNA2 potassium channel endocytosis upon CHRM1 activation. Related references: PMID:9635436 PMID:19403695
- Functions as an allosteric activator of the guanine nucleotide exchange factor ECT2 and may activate phospholipase C epsilon (PLCE1). Related references: PMID:31888991 PMID:16103226
- Subcellular localization: cell membrane, cytoplasm, cytoskeleton, cleavage furrow, midbody, lamellipodium, dendrites, and nucleus.
- Post-translational modifications: prenylation, phosphorylation, methylation, ADP-ribosylation, and ubiquitination. Also subject to proteolytic cleavage by Yersinia pestis yopT during infection.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 94–193 of human RhoA. The antibody is expected to recognize full-length endogenous RhoA, with cross-reactivity predicted in human, mouse, and rat.
- For Western blot (WB), expect a specific band around 22 kDa. Validate detection conditions using appropriate positive and negative controls.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider antigen retrieval techniques and optimize antibody dilution based on your tissue or cell model.
- ELISA applications may require pairing with a matched detection reagent; consider titrating the antibody to determine the optimal coating and detection concentrations.
