Biological Background: RHBDF1 Function and Localization

• RHBDF1 encodes inactive rhomboid protein 1 (iRhom1), a catalytically inactive member of the rhomboid family that functions as a critical regulator of ADAM17 sheddase activity, controlling the release of EGFR ligands and TNF-α. Related references: PMID:15965977 PMID:18524845 PMID:18832597

Experimental Guidance and Technical Tips

• For immunofluorescence/immunocytochemistry (IF/ICC), anticipate ER and Golgi staining patterns. Optimize permeabilization and consider co-staining with organelle markers to confirm subcellular localization.
