Biological Background: RHBDF1 Function and Localization
- RHBDF1 encodes inactive rhomboid protein 1 (iRhom1), a catalytically inactive member of the rhomboid family that functions as a critical regulator of ADAM17 sheddase activity, controlling the release of EGFR ligands and TNF-α. Related references: PMID:15965977 PMID:18524845 PMID:18832597
- It promotes proteasome assembly by facilitating the formation of the PSMG1–PSMG2 chaperone heterodimer, thereby positively regulating proteasome activity. Related references: PMID:26109405
- The protein localizes to the membranes of the endoplasmic reticulum and Golgi apparatus, consistent with its role in protein trafficking and maturation.
- RHBDF1 is highly expressed in the cerebellum, cerebrum, heart, skeletal muscle, placenta, pancreatic islets, and testis; lower expression is detected in colon, kidney, small intestine, and lung.
- It is a multipass transmembrane glycoprotein involved in growth factor binding and intracellular protein transport, as annotated by UniProt keywords.
Experimental Guidance and Technical Tips
- For western blotting (WB), the predicted molecular weight is approximately 97 kDa; however, due to glycosylation, the observed band may migrate at a slightly different position. Use appropriate molecular weight markers and validate with a positive control lysate.
- For immunofluorescence/immunocytochemistry (IF/ICC), anticipate ER and Golgi staining patterns. Optimize permeabilization and consider co-staining with organelle markers to confirm subcellular localization.
- For ELISA, this antibody can be used as either a capture or detection reagent. Titrate the antibody and validate against the immunogen or full-length recombinant protein.
- Cross-reactivity with mouse and rat has been reported; verify sequence homology in the immunogen region if using these species, and perform a pilot experiment to confirm reactivity.
