- Regulator of G protein signaling 4 (RGS4) is a member of the RGS protein family that acts as a negative regulator of G protein-coupled receptor (GPCR) signaling.
- It functions as a GTPase-activating protein (GAP) for heterotrimeric G protein alpha subunits, enhancing GTP hydrolysis to return them to an inactive GDP-bound state.
- RGS4 preferentially modulates G(z)-alpha and G(i)-alpha subtypes; its activity can be inhibited by phosphorylation or palmitoylation of the target G protein alpha subunit.
- The RGS4 gene undergoes alternative splicing, generating multiple isoforms with distinct tissue distributions. Isoforms 4 and 5 are ubiquitously expressed, while isoforms 1, 2, and 3 are absent from the cerebellum.
- RGS4 is predominantly expressed in the brain (prefrontal and visual cortex) and heart at the protein level, aligning with its role in neural and cardiac signaling.
