Biological Background: RGS20 Function and Localization
- RGS20 (Regulator of G protein signaling 20) is a member of the RGS protein family that functions as a GTPase-activating protein (GAP) for heterotrimeric G protein alpha subunits.
- It selectively binds to G(z)-alpha and G(alpha)-i2 subunits, accelerating their intrinsic GTPase activity and thereby driving G proteins into the inactive GDP-bound state, inhibiting signal transduction.
- RGS20 negatively regulates mu-opioid receptor-mediated activation of G proteins (by similarity).
- Its activity is regulated by phosphorylation and palmitoylation of the target G protein.
- The protein exists in multiple isoforms generated by alternative splicing; isoform 5 is specifically expressed in the brain, notably at high levels in the caudate nucleus and temporal lobe.
- Subcellularly, RGS20 localizes to the membrane, nucleus, and cytoplasm.
- Post-translational modifications include glycosylation, lipidation (palmitoylation), phosphorylation, and ubiquitin-like conjugation (Ubl conjugation), reflecting its dynamic regulation.
Experimental Guidance and Technical Tips
- For Western blot, the predicted molecular weight is ~44kDa; consider using appropriate tissue or cell lysates expressing RGS20, such as brain tissues, especially caudate nucleus or temporal lobe regions, based on isoform-specific expression.
- For ELISA, the recombinant immunogen can serve as a positive control to validate antibody binding.
- Since the antibody cross-reacts with mouse and rat, it is suitable for studies in rodent models; however, verify specificity in the relevant sample system.
- Given the polyclonal nature and immunogen design (full N-terminal fragment), the antibody may recognize multiple isoforms; reviewing the target sequence alignment across species and isoforms is advisable.
